I. Background | |
毒蕈堿乙酰受體(MAChRs,簡稱 M 受體)參與中樞神經(jīng)系統(tǒng)的運動控制、體溫 調(diào)節(jié)、心血管調(diào)節(jié)和記憶,介導周圍神經(jīng)系統(tǒng)的平滑肌收縮、腺體分泌和心率、調(diào)節(jié)心肌 收縮力。CHRM2 屬于 GPCR,是 MAChRs 的 5 種亞型之一,可以與 Gi/o 偶聯(lián)。CHRM2 是心臟和腦干中最主要的毒蕈堿受體亞型。CHRM2 通過 Gi/o 信號精密抑制心臟與神經(jīng)元 過度活動,維持內(nèi)環(huán)境穩(wěn)態(tài)。其功能失衡直接導致心動過緩、癲癇易感性升高及抗抑郁治 療抵抗,是神經(jīng)心血管藥物設(shè)計的核心靶標。 | |
