信使RNA(mRNA)5 '帽狀結(jié)構(gòu)能夠防止mRNA在細(xì)胞質(zhì)中的降解,促進(jìn)核糖體增長(zhǎng)和蛋白的翻譯。Trilink的帽類似物能夠?qū)崿F(xiàn)共轉(zhuǎn)錄加帽,這樣就省去了加帽酶加帽操作。
CleanCap AG是TriLink公司用于5 '共轉(zhuǎn)錄加帽的產(chǎn)品,CleancapAG加帽可以形成Cap 1結(jié)構(gòu)。CleancapAG的加帽效率高達(dá)98%。請(qǐng)注意:TriLink建議盡可能使用CleanCap AG,模板應(yīng)該使用配套的AG啟動(dòng)子。Cleancap AG的加帽效率通常>90%。不像CleanCap AG,使用CleanCap GG(貨號(hào)cat#N-7133或cat#N-7433)可能導(dǎo)致類似的ARCA轉(zhuǎn)錄效率和產(chǎn)量下降的現(xiàn)象。

Cleancap AU是mRNA表達(dá)過(guò)程中用于共轉(zhuǎn)錄加帽的產(chǎn)品,能夠同步生產(chǎn)出具有天然結(jié)構(gòu)Cap 1的mRNA。CleanCapAU是專為自復(fù)制RNA(saRNA)設(shè)計(jì)的,該saRNA基于正義RNA病毒,如委內(nèi)瑞拉馬腦炎病毒(VEEV)、塞姆利基森林病毒(SFV)和辛德比斯病毒(SIN)的基因組。這些正義RNA病毒基因組起始是一個(gè)5'-AU…Cap 1 mRNA。與傳統(tǒng)的共轉(zhuǎn)錄轉(zhuǎn)錄加帽方法產(chǎn)生的Cap 0 mRNA相比,具有更高的體內(nèi)活性。CleanCap可以與TriLink的修飾的NTP產(chǎn)品一起使用。
